Atomic-resolution Structure of Cyanobactrial Cytochrome C6 with an Unusual Sequence Insertion

چکیده
During photosynthesis, electron transfer between two membrane-bound complexes, cytochrome b6f and photosystem I can be accomplished by the coppercontaining protein, plastocyanin, or the heme protein, cytochrome c6 [1]. Cytochromes c6 are water-soluble, low-spin heme-containing proteins involved in the highpotential (340-390 mV) electron transport chain. They are characterized by low molecular mass, 80-90 amino acid residues in the mature protein, and have a covalently bound heme group. Cytochrome c6 from the mesophilic cyanobacterium Synechococcus sp. PCC 7002 is unique among all known cytochromes c6 due to the presence of an unusual sevenresidue insertion, K44DGSKSL50. Furthermore, the present protein is unusual because of its very high content (36%) of the smallest residues (glycine and alanine) [2]. The crystallization experiments used the hangingdrop vapor-diffusion method. Red crystals (Fig. 1) suitable for X-ray analysis were obtained from 10 mM sodium Hepes, pH 6.2, and 2.2 M ammonium sulfate over a period of one week.

Structural Characteristics of Stable Folding Intermediates of Yeast Iso-1-Cytochrome-c
Cytochrome-c (cyt-c) is an electron transport protein, and it is present throughout the evolution. More than 280 sequences have been reported in the protein sequence database (www.uniprot.org). Though sequentially diverse, cyt-c has essentially retained its tertiary structure or fold. Thus a vast data set of varied sequences with retention of similar structure and fun...
متن کاملThe novel cytochrome c6 of chloroplasts: a case of evolutionary bricolage?
Cytochrome c6 has long been known as a redox carrier of the thylakoid lumen of cyanobacteria and some eukaryotic algae that can substitute for plastocyanin in electron transfer. Until recently, it was widely accepted that land plants lack a cytochrome c6. However, a homologue of the protein has now been identified in several plant species together with an additional isoform in the green alga Ch...
متن کاملAn atomic-resolution view of neofunctionalization in the evolution of apicomplexan lactate dehydrogenases
Malate and lactate dehydrogenases (MDH and LDH) are homologous, core metabolic enzymes that share a fold and catalytic mechanism yet possess strict specificity for their substrates. In the Apicomplexa, convergent evolution of an unusual LDH from MDH produced a difference in specificity exceeding 12 orders of magnitude. The mechanisms responsible for this extraordinary functional shift are curre...
متن کاملAn Unusual Cause of Cervical Radicular Pain-Foreign Body in Esophagus
Introduction: Foreign bodies in the esophagus are considered to be a life-threatening condition in adults and children because of esophageal perforation, chemical pneumonitis, airway obstruction, and development of a fistula, leading to high morbidity and mortality with this condition. Most cases present with immediate symptoms. However, in rare cases, the foreign body can migrate within the t...
متن کاملAn investigation of neutron direct damages at energies of 0.1-2 MeV on the DNA molecules with atomic structure deduced using Geant4 toolkit
This study proposes a method to estimate RBE of fast neutrons using Monte Carlo simulations. This approach is based on the combination of an atomic resolution DNA geometrical model and Monte Carlo simulations for tracking particles. Atomic positions were extracted from the Protein Data Bank. The GEANT4 code was used for tracking the secondary particles generated by fast neutrons during their in...
